Function |
Substrate-binding subunit of the Rab geranylgeranyltransferase (GGTase) complex. Binds unprenylated Rab proteins and presents the substrate peptide to the catalytic component B composed of RABGGTA and RABGGTB, and remains bound to it after the geranylgeranyl transfer reaction. The component A is thought to be regenerated by transferring its prenylated Rab back to the donor membrane. Besides, a pre-formed complex consisting of CHM and the Rab GGTase dimer (RGGT or component B) can bind to and prenylate Rab proteins; this alternative pathway is proposed to be the predominant pathway for Rab protein geranylgeranylation.

Summary |
This gene encodes component A of the RAB geranylgeranyltransferase holoenzyme. In the dimeric holoenzyme, this subunitbinds unprenylated Rab GTPases and then presents them to thecatalytic Rab GGTase subunit for the geranylgeranyl transferreaction. Rab GTPases need to be geranylgeranyled on either one ortwo cysteine residues in their C-terminus to localize to thecorrect intracellular membrane. Mutations in this gene are a causeof choroideremia; also known as tapetochoroidal dystrophy (TCD).This X-linked disease is characterized by progressive dystrophy ofthe choroid, retinal pigment epithelium and retina. Alternativesplicing results in multiple transcript variants encoding differentisoforms.
