Function	Converts sphingomyelin to ceramide. Also has phospholipase C activities toward 1,2-diacylglycerolphosphocholine and 1,2-diacylglycerolphosphoglycerol. Isoform 2 and isoform 3 have lost catalytic activity.
Summary	The protein encoded by this gene is a lysosomal acid sphingomyelinase that converts sphingomyelin to ceramide. The encoded protein also has phospholipase C activity. Defects in this gene are a cause of Niemann-Pick disease type A (NPA) and Niemann-Pick disease type B (NPB). Multiple transcript variants encoding different isoforms have been identified. Images	Overlay histogram showing K562 cells stained with APR07182G (green line). The cells were fixed with 2% paraformaldehyde (10 min) and then permeabilized with 90% methanol for 10 min. The cells were then icubated in 2% bovine serum albumin to block non-specific protein-protein interactions followed by the antibody (APR07182G, 1:25 dilution) for 60 min at 37?C. The secondary antibody used was Goat-Anti--Rabbit IgG, DyLight? 488 Conjugated Highly Cross-Adsorbed(OH191631) at 1/200 dilution for 40 min at 37?C. Isotype control antibody (blue line) was rabbit IgG (1μg/1x10^6 cells) used under the same conditions. Acquisition of >10, 000 events was performed. All lanes : Anti-SMPD1 Antibody (S376) at 1:2000 dilution Lane 1: human cerebellum lysate Lane 2: HepG2 whole cell lysate Lane 3: K562 whole cell lysate Lane 4: MCF-7 whole cell lysate Lysates/proteins at 20 μg per lane. Secondary Goat Anti-Rabbit IgG, (H+L), Peroxidase conjugated at 1/10000 dilution. Predicted band size : 70 kDa Blocking/Dilution buffer: 5% NFDM/TBST. SMPD1 Antibody (C-term) (Cat. APR07182G)immunohistochemistry analysis in formalin fixed and paraffin embedded human testis tissue followed by peroxidase conjugation of the secondary antibody and DAB staining.This data demonstrates the use of SMPD1 Antibody (C-term) for immunohistochemistry. Clinical relevance has not been evaluated.
