The proteasome is a multicatalytic proteinase complex which is characterized by its ability to cleave peptides with Arg, Phe, Tyr, Leu, and Glu adjacent to the leaving group at neutral or slightly basic pH. The proteasome has an ATP-dependent proteolytic activity. This unit is responsible of the chymotrypsin-like activity of the proteasome and is one of the principal target of the proteasome inhibitor bortezomib. May catalyze basal processing of intracellular antigens. Plays a role in the protection against oxidative damage through the Nrf2-ARE pathway (By similarity).

Summary |
The proteasome is a multicatalytic proteinase complex witha highly ordered ring-shaped 20S core structure. The core structureis composed of 4 rings of 28 non-identical subunits; 2 rings arecomposed of 7 alpha subunits and 2 rings are composed of 7 betasubunits. Proteasomes are distributed throughout eukaryotic cellsat a high concentration and cleave peptides in anATP/ubiquitin-dependent process in a non-lysosomal pathway. Anessential function of a modified proteasome, the immunoproteasome,is the processing of class I MHC peptides. This gene encodes amember of the proteasome B-type family, also known as the T1Bfamily, that is a 20S core beta subunit in the proteasome. Thiscatalytic subunit is not present in the immunoproteasome and isreplaced by catalytic subunit 3i (proteasome beta 8 subunit).Multiple transcript variants encoding different isoforms have beenfound for this gene.
