Function |
Transcriptional activator required for lipid homeostasis. Regulates transcription of the LDL receptor gene as well as the cholesterol and to a lesser degree the fatty acid synthesis pathway (By similarity). Binds the sterol regulatory element 1 (SRE-1) (5'-ATCACCCCAC-3') found in the flanking region of the LDRL and HMG-CoA synthase genes.

Summary |
SREBP2 regulates cholesterol synthesis by activating the transcription of genes for HMG-CoA reductase and other enzymes of the cholesterol synthetic pathway.2 SREBP2 is ubiquitously detected in various tissues. Under basal conditions SREBP is bound to ER membranes as a glycosylated precursor protein. Upon cholesterol depletion, the protein is cleaved to its active forms (about 50-68 kDa) and translocated into the nucleus to stimulate transcription of genes involved in the uptake and synthesis of cholesterol.

Flurescent image of U251 cell stained with SREBP2 Antibody. Cells were fixed with 4% PFA(20min), permeabilized with Triton X-100 (0.1%, 10min), then incubated with SREBP2 primary antibody (1:25, 1h at 37℃). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:400, 50min at 37℃). Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (7units/ml, 1h at 37℃). SREBP2 immunoreactivity is localized to Nucleus and Nucleolus siginficantly.
SREBF2 Antibody western blot analysis in HepG2 cell line lysates (35ug/lane). This demonstrates the SREBP2 antibody detected the SREBP2 protein.
SREBP2 antibody immunohistochemistry analysis in formalin fixed and paraffin embedded human skin carcinoma followed by peroxidase conjugation of the secondary antibody and DAB staining. This data demonstrates the use of the SREBP2 antibody for immuohistochemistry. Clinical relevance has not been evaluated. |
