Signal transducer and transcription activator that mediates cellular responses to interleukins, KITLG/SCF and other growth factors. May mediate cellular responses to activated FGFR1, FGFR2, FGFR3 and FGFR4. Binds to the interleukin-6 (IL-6)- responsive elements identified in the promoters of various acute- phase protein genes. Activated by IL31 through IL31RA. Cytoplasmic STAT3 represses macroautophagy by inhibiting EIF2AK2/PKR activity. Plays an important role in host defense in methicillin-resistant S.aureus lung infection by regulating the expression of the antimicrobial lectin REG3G (By similarity).

Summary

STAT3 is a member of the STAT protein family. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein is activated through phosphorylation in response to various cytokines and growth factors including IFNs, EGF, IL5, IL6, HGF, LIF and BMP2. It mediates the expression of a variety of genes in response to cell stimuli, and thus plays a key role in many cellular processes such as cell growth and apoptosis. The small GTPase Rac1 has been shown to bind and regulate the activity of this protein. PIAS3 protein is a specific inhibitor of this protein.

Form

Purified polyclonal antibody supplied in PBS with 0.09% (W/V) sodium azide. This antibody is purified through a protein A column, followed by peptide affinity purification.

Fluorescent confocal image of SY5Y cells stained with phospho-STAT3-S727 antibody. SY5Y cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with APR11163G phospho-STAT3-S727 primary antibody (1:200, 2 h at room temperature). For secondary antibody, Alexa Fluor? 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 μg/ml, 5 min). Note the highly specific localization of the phospho-STAT3 immunosignal mainly to the nucleus, supported by Human Protein Atlas Data (http://www.proteinatlas.org/ENSG00000168610).

The Phospho-STAT3-S727 Pab (Cat. 3260a) is used in western blot to detect Ser727-phosphorylated STAT3 in mouse liver tissue lysates collected before (-) or after (+) stimulation obtained by IL-6 injection in mouse portal vein. Data is kindly provided by Drs. E. Bard-Chapeau and G-S. Feng from the Burnham Institute (La Jolla, CA).

The phospho-STAT3-Ser727 Pab (Cat. APR11163G) is used in Western blot to detect Ser727-phosphorylated STAT3 in HL-60 tissue lysates collected before (-) or after (+) stimulation obtained by IFN-a.
