Recognizes the DNA sequence 5'-ATTAA-3'. Transcriptional repressor. May play a role in hematopoietic differentiation. Establishes anterior identity at two levels; acts early to enhance canonical WNT-signaling by repressing expression of TLE4, and acts later to inhibit NODAL-signaling by directly targeting NODAL (By similarity).

Summary |
Hex, the hematopoietically-expressed homeobox protein (proline-rich homeodomain protein/PRH) is a transcription factor that is important in the control of cell proliferation and differentiation and in the regulation of multiple processes in embryonic development. Hex contains two domains that can independently bring about transcriptional repression. The Hex homeodomain represses transcription by binding to TATA box sequences, whereas the proline-rich N-terminal domain of Hex can repress transcription when attached to a heterologous DNA-binding domain (1). Hex is expressed in primitive endoderm, in some endoderm-derived tissues and in endothelial and hematopoietic precursors. Expression is extinguished during terminal differentiation of endothelial and hematopoietic cells as well as in adult lung (2).
