Summary

There are at least four distinct but related alkalinephosphatases: intestinal, placental, placental-like, andliver/bone/kidney (tissue non-specific). The first three arelocated together on chromosome 2 while the tissue non-specific formis located on chromosome 1. The product of this gene is a membranebound glycosylated enzyme, also referred to as the heat stableform, that is expressed primarily in the placenta although it isclosely related to the intestinal form of the enzyme as well as tothe placental-like form. The coding sequence for this form ofalkaline phosphatase is unique in that the 3' untranslated regioncontains multiple copies of an Alu family repeat. In addition, thisgene is polymorphic and three common alleles (type 1, type 2 andtype 3) for this form of alkaline phosphatase have been wellcharacterized.

Storage & Stability

Store at +4°C short term. For long-term storage, aliquot and store at -20°C or below. Stable for 12 months at -20°C. Avoid repeated freeze-thaw cycles.

Images

Fluorescent confocal image of HeLa cells stained with ALPP (N-term) antibody. HeLa cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with AMM02400G ALPP (N-term) primary antibody (1:100, 2 h at room temperature). For secondary antibody, Alexa Fluor? 488 conjugated donkey anti-mouse antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 μg/ml, 5 min). ALPP immunoreactivity is localized to the cytoplasm of HeLa cells.

ALPP Antibody (N-term) (Cat. AMM02400G) western blot analysis in mouse placenta tissue lysates (35μg/lane).This demonstrates the ALPP antibody detected the ALPP protein (arrow).
