Coronavirus SARS-CoV-2 (2019-nCoV) Antigen Reagent

ICTV (International Committee on Taxonomy of Viruses) announced “severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)” as the name of the new virus on 11 February 2020. This name was chosen because the virus is genetically related to the coronavirus responsible for the SARS outbreak in 2003. WHO officially announced “COVID-19” as the name of this new disease on 11 February 2020, following guidelines previously developed with the World Organisation for Animal Health (OIE) and the Food and Agriculture Organization of the United Nations (FAO).


The coronavirus genome encodes four or five structural proteins. They are a spike protein [S], a small envelope protein [E], a membrane protein [M], a nucleocapsid protein [N], and sometimes a hemagglutinin-esterase protein [HE]. 


The spike protein is the major surface membrane protein of coronavirus, which is composed of two subunits, S1 and S2. S1 can function as a receptor-binding domain (RBD) while S2 contains the basic elements required for the membrane fusion process.


The Spike protein is responsible for binding to the host cell receptor and membrane fusion. It plays key parts in neutralizing host antibodies and vaccine design. The interaction between Spike protein of SARS-CoV-2 and RBD-human ACE2 interfaces to enhance the viral binding to human ACE2 and plays a critical role in the infection of respiratory epithelial cells.


The nucleocapsid protein is one of the most abundant structural proteins during SARS-CoV infection and is highly phosphorylated. During the assembly of the virion, the N protein binds to the viral RNA and causes the formation of a spiral nucleocapsid. Nucleocapsid protein is a highly immunogenic phosphoprotein, which is involved in viral genome replication and regulating cell signaling pathways. Due to the sequence conservation and strong immunogenicity of N protein, N protein is often used as a diagnostic tool for coronavirus.

Angiotensin-converting enzyme 2 (ACE2)

Angiotensin-converting enzyme 2 (ACE2) is a membrane receptor expressed on the surface of airway epithelial cells. In the early 21st century, scientists began to conduct in-depth research on ACE2. ACE2 has been shown to be the host cell receptor for two human coronaviruses, SARS-COV and HCoV-NL63. And it can interact with the coronavirus Spike protein. This interaction which mediated by the RBD domain of the Spike protein is considered to be a key step in membrane fusion between the virus and the cell. Recently, ACE2 has once again attracted great attention for it is shown to be a host cell receptor for a newly discovered lethal coronavirus SARS-CoV-2 (2019-nCoV). This finding suggests that blocking the interaction between S protein and ACE2 is an effective treatment for coronavirus infection.


Theoretically, both SARS-CoV-2 virus and receptor ACE2 can be used as therapeutic targets for effective intervention against viral infection. Soluble Spike proteins or blocking antibodies can be used to block ACE2 to prevent viral infection. Similarly, soluble ACE2-Fc fusion protein can be used to block S1 protein on virions to achieve the same effect.


In addition to its important role in mediating virus entry into host cells, ACE2 is very likely to play a pivotal role in the pathogenesis of respiratory diseases. ACE2 is an important regulatory factor of the renin-angiotensin system (RAS). ACE2 can negatively regulates its activity by converting angiotensin II into the product angiotensin 1-7. Dysregulation of ACE2 can be observed in lung lesions caused by RSV and avian influenza (H5N1) infection. And it is also shown that the use of ACE2-Fc fusion protein can reduce symptoms in animal models.
