If you are sure you can see mutant protein being expressed even at low concentration, according to me best would be to do a western blotting thats the prefect way to confirm the expressed protein. otherwise you can also cut out the band from gel and send for LC/MS-MS to detect the expressed protein. Personally if i know my protein is being expressed , I wont spend lot of time in optimisation as takes lot of time. rather than I will try to prove that protein is being expressed if thats the main aim.
