Why do some proteins run with high molecular weight?

Date: 2020-08-11 Author: Leading Biology Click: 500

A protein my run at a higher molecular weight for a number of reasons:

(1) It has been post-translationally modified, especially by glycosylation, which increased its mass.

(2) It was not fully denatured prior to electrophoresis. Some proteins are very stable to denaturation.

(3) It has an unusually high content of basic amino acids.

(4) It was not fully reduced and retains one or more disulfide bonds.
