How were proteins discovered and purified before sequencing?

Date: 2020-08-10 Author: Leading Biology Click: 490

Through purification of proteins, we can determine amino acid sequences and evolutionary relationships between proteins in diverse organisms.

Purification should yield a sample of protein containing only one type of molecule.

To start the purification, we need to decide the amount of protein present in the mixture being assayed. With these two experimentally determined numbers—enzyme activity and protein concentration—we then calculate the specific activity, the ratio of enzyme activity to the amount of protein in the enzyme assay. The specific activity will rise as the purification proceeds and the protein mixture being assayed consists of a greater and greater extent of lactate dehydrogenase.

Then Proteins is released from the Cell to Be Purified.
