Why is one primer used for DNA sequencing instead of two?

Date: 2020-07-29 Author: Leading Biology Click: 483

Excess PCR primers and unincorporated nucleotides should be removed before sequencing is started. Since one primer is used in each sequencing reaction, while 2 primers are used in PCR, if the 2 primers are not removed in PCR, you will get two sequences superimposed on each other that are not readable.
