Template Preparation: high purity template has a great effect on the success of automated sequencing. There are many commercial kits available. I recommend using Leading Biology PCR kits for sequencing since the yield could be ensured. In addition to the DNA in the amount of at least 2μg is needed, it’s suggested to prepare an extra amount of DNA in case that the first reaction fails, we still have enough samples for a re-sequencing and the experiment will not be delayed. Be sure that the DNA must be free of contaminants such as nucleases, salts, nucleotides, and ethanol.
