DNA double-stranded fragmentation enzyme is a time-dependent enzyme, which can cut dsDNA into 50-1, 000 bp fragments according to different time of action. DNA double-stranded fragmenting enzyme is composed of two enzymes. One enzyme randomly generates the cutting on dsDNA, and the other enzyme recognizes the cutting point and performs the cutting on the chain to generate the dsDNA fragment. The resulting DNA fragment contains very short protruding ends, 5 acids and 3 acids containing hydroxyl groups. After library preparation and sequencing verification, DNA double-stranded fragment enzyme has the characteristics of random cutting. Compared with the genomic DNA libraries prepared by mechanical method, the sequencing results showed no bias and no difference in sequencing coverage.
