Interference Function and Anti-idiotype Antibody ServiceIntroduction
Although a wide variety of antigens in the immune animal could provide a wide variety of antibodies, even the same antigen processed by immune cells could present different antigen information to B lymphocytes, so the pairing relationship between antigens and antibodies is mainly multiple to one, but in converse, for a specific antibody, there is only one antigen or antigen information could be identified. Among the hundreds of amino acids that constitute an antibody, most of the sequences are highly conserved in the same antibody subtype family. Only a few of them which closely related to antigen binding are different, and these amino acid sequences are unique sequences identifying specific antigen.
In many medical or biological studies, it is often needed to do qualitative and quantitative analyses for the specific antibodies induced by antigens (such as a drug), in order to get some indicator information (such as some pharmacokinetics indicators). If there is no specific anti-antibodies to identify specific antibodies, the immunoglobulin family derived from the target antibody will cause great interference to the experiment, even submerged the meaningful experimental data in the background. And as for some antibody which itself is an antigen (such as antibody drug), it is often difficult to effectively trigger the immune response, and to distinguish these antibodies and antibody form the body itself, to achieve in situ or real-time detection, we also need specific anti-antibodies which could identify specific antibody.
Fig. 1 For an antibody, the specific binding site of antigen is also the core location for the antibody identification, the epitope of a specific antibody is usually located in this region.
On the one hand, the specificity of the antibody ensures the specificity of the recognition, on the other hand, these antibodies often accompanied by repression function, the target antigen (i.e. the antibody which be identified, referred as TA) combination could block the original antigen (i.e. the antigen which identified by TA, also recognized by primary antigen, PA) binding.
Due to the unique function mentioned above, these specific anti-antibodies are often used in the study of pharmacokinetics (PK).

A. The normal combination of the secondary antibody and primary antibody only have species specificity, but no specificity of antibody recognition. It can be used in the PK study in the form of antigen capture.
B. Preparation of antigen-antibody complex antibody can be used in PK study of conjugated drugs, this antibody only recognizes antibodies which in binding state, thus has high specificity for drug binding recognition, but the preparation is difficult, and it also have high requirement of the TAb binding force and conformation stability.
C. The most commonly binding form used in the PK study of antibody drugs (AD) or anti-drug antibodies (ADA).
D. Binding specific anti-antibody preparation is based on C, by using capture-targeting-reporting conjugation method, detect target antibody directly in the original habitat, according to the experimental purpose and PK experimental design, there are different experimental requirements for two antibodies specification.
